It could be if you can send a screen shot of what repetier looks like we may be able to see what is wrong and then go from there

By the way where you located ?

I am located in The Netherlands.

Later today (our time) I'll try again and make a screen shot. Most of time the software is waiting for temperature of the extruder and bed, because I can not set the temperature as necessary.

It sounds like you may have connected but are still not communicating properly that is most likely what is happening on the right side do you see anything like 5 commands waiting??

    No.

Sometimes it works - sometimes not.

What is the best point of time to connect (Repetier) with the printer? Before of once a template is loaded or at the end? Or this does not matter?

USB devices are found when powered on or connected so it should not matter I leave mine on 24/7

Also Make sure you are connecting directly not through a hub or extension

Sure! I have the origin cable.

With the best point of time to connect, I meant (after Repetier is started): immediately click connect or only before I start to print?

I hook up the cable and power on the printer and click connect and just leave it that way until I power off you should be able to do it anytime

Ready with the first complete print! I am proud...but it looks like a little amateurish.

Now I am trying to do my second print. A logo of my car (Infinity). Connection exist but the bed temperature is going down and will remain low. I cancel this now and try again.

The connection problems still exists: reset printer, usb cabel out and in, and so on. After a few trials Repetier get connected with the printer.

    Now I've got this message:

< 21:40:07: Error:printer stopped due to errors. Fix the error and use M999 to restart. (Temperature is reset. Set it after restarting)

    what means M999?

Try moving the two yellow fuses on the RAMPS board apart from one another and give them plenty of space between. Also, check your connection on the RAMPS to make sure that the wires are snugged down tight.
