I was replacing the Extruder for the E3D v6 and I lost one of the bolts that hold the extruder in place. I tried to get some from home depot but I did find the right size . Does anyone know the size and threading or an eBay page with a replacement size

exactly what printer are we talking about?
There are several models and the way things are attached is different.

The Beta and R1 (but not the R1+) used M3-25mm screws to hold the extruder in place.

Are you sure? Do you have couplers on your threaded rods for z? Is the hotend held in place with 2 screws from the side or two screws from the bottom through a "quick release" plate that you pull out to remove the hotend?

The screws come in from the side and couplers are on the z

Should be fine. Some of the early ones came with Phillips head screws and those were terrible. The Hex head screws are far better.
